Due to their genetically encoded nature, peptides are promising candidates for new antimicrobial drugs. Lantipeptide andalusicin exhibits significant antimicrobial activity against Gram-positive bacteria, making it a promising scaffold for the development of DNA-encoded libraries of lantibiotics. In this study, the modification reaction of andalusicin by class III lanthionine synthetase AncKC was reconstructed in a heterologous <italic>Escherichia</italic><italic> </italic><italic>coli</italic> system. The results obtained open possibilities for creating novel peptide-based antimicrobial agents.</p></abstract><trans-abstract xml:lang="ru"><p>Возрастающая устойчивость микроорганизмов к антибиотикам требует поиска новых антимикробных средств. Пептиды, благодаря своей генетически кодируемой природе, являются перспективными кандидатами на роль новых антимикробных препаратов. Лантипептид андалусицин обладает выраженной антимикробной активностью против грамположительных бактерий, поэтому он может рассматриваться как перспективная матрица для создания ДНК-кодируемых библиотек лантибиотиков. Проведена реконструкция реакции модификации андалусицина лантионинсинтетазой III класса AncKC в гетерологической системе <italic>Escherichia с</italic><italic>oli</italic>.
