This enzyme has a broad spectrum of efficacy against organophosphorus compounds, which makes it a prime candidate for the role of stoichiometric bioscavenger. Development of a new-age DNA-encoded bioscavenger is a vival task. Several transgenic expression systems of human BChE were developed over the past 20 years; however, none of them has been shown to make economic sense or has been approved for administration to humans. In this study, a CHO-based expression system was redesigned, resulting in a significant increase in the production level of functional recombinant human butyrylcholinesterase as compared to the hitherto existing systems. The recombinant enzyme was characterized with Elman and ELISA methods.</p>
В представленной работе разработана эффективная система экспрессии рекомбинантной бутирилхолинэстеразы человека на основе клеток линии CHO, а также охарактеризован белок, полученный с использованием этой системы.</p></trans-abstract><kwd-group xml:lang="en"><kwd>bioscavenger</kwd><kwd>butyrylcholinesterase</kwd><kwd>CHO cell line</kwd><kwd>recombinant protein</kwd><kwd>оrganophosphorus toxins</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>биологический антидот</kwd><kwd>бутирилхолинэстераза</kwd><kwd>клетки линии CHO</kwd><kwd>рекомбинантный белок</kwd><kwd>фосфорорганические токсины</kwd></kwd-group><funding-group><funding-statement xml:lang="en">This work was supported by the Russian Foundation for Basic Research (grant No. 10-04-00673-a), the Federal Target-oriented Program (No. 2046.2012.4) of the Scientific School “The Chemical Foundations of Biocatalysis”, the Program of the Presidium of RAS No. 24 “Nanotechnologies and Nanomaterials,” and the Foundation for assistance to small enterprises in the field of science and technology (Program ”U.M.N.I.K.”).</funding-statement><funding-statement xml:lang="ru">Работа поддержана РФФИ (грант № 10-04-00673-а), ФЦП (государственный контракт № 2046.2012.4) научной школы «Химические основы биокатализа», Программой Президиума РАН № 24 «Нанотехнологии и наноматериалы» и Фондом содействия развитию малых форм предприятий в научно-технической сфере - программа «УМНИК».</funding-statement></funding-group></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>1.
