Dimerization ability has been shown for several ZAD-domains. The functional role of this domain is poorly understood. In this paper, we demonstrate that a point mutation within the ZAD-domain of the Zw5 insulator protein disrupts its nuclear localization without affecting its dimerization ability. The importance of the ZAD-domain for nuclear localization has also been shown for the Pita and Grauzone proteins. Therefore, one of the ZAD-domain functions is control of the nuclear localization of transcription factors.</p></abstract><trans-abstract xml:lang="ru"><p>Многие транскрипционные факторы с мотивами цинковых пальцев типа С2Н2 содержат на N-конце характерный домен, названный ZAD (Zinc-finger Associated Domain), в котором четыре остатка цистеина координируют ион цинка. Показано, что некоторые ZAD-домены обладают способностью к гомодимеризации. Однако функциональная роль этого домена остается почти неизученной. Нами показано, что точечная мутация в ZAD-домене инсуляторного белка Zw5 нарушает его ядерную локализацию, не влияя на способность к димеризации. Установлено также, что ZAD-домены необходимы для ядерной локализации белков Pita и Grau.
